Recombinant NY-ESO-1 protein is a cancer vaccine component derived from the NY-ESO-1 gene (cancer/testis antigen 1B). It is an 18-kDa protein with 180 amino acids featuring a glycine-rich N-terminal region and a hydrophobic C-terminal region containing a conserved Pcc-1 domain. It functions as both a tumor-associated antigen and its own adjuvant. When administered, particularly with adjuvants like ISCOMATRIX, it induces broad integrated antibody and CD4+ and CD8+ T cell responses against NY-ESO-1 expressing tumors. Clinical trials have shown that recombinant NY-ESO-1 protein is well-tolerated and highly immunogenic, generating high-titer antibody responses, strong delayed-type hypersensitivity reactions, and T cells specific for a broad range of NY-ESO-1 epitopes. The protein's expression is predominantly restricted to various solid tumors, germ cells, and placental cells, with little to no expression in normal adult somatic tissues. This restricted expression pattern makes NY-ESO-1 a target with limited off-target toxicities and broad applications in numerous cancer types.
