Overview

Recombinant paraoxonase 1 (rHuPON1) is a biologic therapeutic enzyme being developed as a catalytic bioscavenger for the treatment and prophylaxis of organophosphate poisoning, including exposure to nerve agents and insecticides. It is a calcium-dependent hydrolytic enzyme naturally associated with high-density lipoproteins (HDL) in blood plasma. The enzyme functions by hydrolyzing and detoxifying neurotoxic organophosphates and insecticide metabolites. Beyond its role as a bioscavenger, rHuPON1 has potential applications in cardiovascular disease due to its ability to hydrolyze oxidized phospholipids and in oncology, specifically hepatocellular carcinoma, where it may act as a metabolic regulator to counteract immunosuppression by promoting the degradation of HIF-1α.
