Drug intelligence / Profile preview
Recombinant PEX5 protein (rpPEX5) is a recombinant form of the peroxisomal biogenesis factor 5 (PEX5), which acts as a cytosolic receptor for peroxisomal matrix protein import. Beyond its established intracellular role in peroxisomal protein trafficking, extracellular recombinant PEX5 has been shown to act as a negative regulator of osteoclastogenesis. It inhibits receptor activator of nuclear factor kappa-B ligand (RANKL)-induced osteoclast differentiation and bone resorption by downregulating NFATc1 and dampening RANKL-mediated signaling pathways (including Akt, MAPK, IκB, and calcium-dependent pathways). In preclinical models, administration of recombinant PEX5 protein mitigates lipopolysaccharide (LPS)-induced inflammatory calvarial bone destruction, suggesting potential therapeutic utility in osteolytic diseases.
