Recombinant RSPO3 (rRSPO3) is a recombinant protein version of R-spondin 3, a member of the R-spondin family that acts as a potent potentiator of the Wnt/β-catenin signaling pathway. In colorectal cancer (CRC), RSPO3 expression is frequently downregulated, and its low expression is associated with poor prognosis and reduced infiltration of immune cells such as dendritic cells (DCs), NK cells, and CD8+ T cells. Research presented at ASCO 2025 indicates that restoration of RSPO3 expression or exogenous treatment with rRSPO3 inhibits CRC tumor growth in vivo by activating the immune response within the tumor microenvironment. Specifically, rRSPO3 treatment has been shown to increase the infiltration of various immune cells, particularly DCs. Furthermore, rRSPO3 exhibits synergistic inhibitory effects when combined with anti-PD-1 therapy in mouse models, suggesting its potential as a therapeutic target for CRC immunotherapy.
