The recombinant SARS-CoV deltaTM spike protein vaccine is an investigational subunit vaccine candidate designed to elicit protective immunity against the Severe Acute Respiratory Syndrome coronavirus (SARS-CoV), which emerged in 2003. The vaccine utilizes a recombinant form of the viral spike (S) glycoprotein that has been engineered to lack the transmembrane (TM) and cytoplasmic domains (denoted as deltaTM or ΔTM). This modification results in a soluble protein that is secreted during production but retains the critical S1 and S2 subunits, including the receptor-binding domain (RBD). By presenting these viral epitopes to the immune system, the vaccine induces the production of neutralizing antibodies that block the interaction between the viral S protein and the host cell receptor, angiotensin-converting enzyme 2 (ACE2). Primarily developed by the National Institute of Allergy and Infectious Diseases (NIAID) and academic partners, research on this candidate provided a foundational understanding of coronavirus immunology, although clinical development for the original SARS-CoV was discontinued following the eradication of the virus from human populations.
