Overview

Recombinant vaccinia-B7.1 vaccine is a genetically engineered cancer immunotherapy that uses a modified vaccinia virus to express the human B7.1 (CD80) costimulatory molecule[2][5][6]. The primary mechanism involves enhancing T-cell mediated immune responses against tumor cells by increasing local expression of B7.1 within the tumor microenvironment, thereby overcoming local T cell tolerance and promoting anti-tumor immunity[2][5][6]. This approach has been investigated primarily in patients with metastatic melanoma through intralesional administration of the vaccine into accessible tumors[4][5]. Clinical studies have shown that this therapy can induce objective tumor regression or disease stabilization in some patients with advanced melanoma, and it is generally well tolerated with mild side effects such as low-grade fever and fatigue[5][6].
