Overview

Recombinant vaccinia-TRICOM vaccine is a therapeutic cancer vaccine that uses a genetically engineered (recombinant) vaccinia virus as a vector to deliver genes encoding three human T-cell costimulatory molecules—B7.1 (CD80), ICAM-1 (CD54), and LFA-3 (CD58)—collectively known as TRICOM. The vector can also be engineered to express tumor-associated antigens such as carcinoembryonic antigen (CEA) or prostate-specific antigen (PSA). The addition of these costimulatory molecules enhances the activation of cytotoxic T lymphocytes against tumor cells expressing the target antigen, thereby stimulating an anti-tumor immune response. This approach aims to improve immunogenicity and clinical efficacy compared to vaccines lacking these co-stimulatory signals[5][6][9]. Recombinant vaccinia-TRICOM vaccines have been studied in various solid tumors, including colorectal cancer and prostate cancer[1][2][5].
