Overview

Recombinant vaccinia virus encoding mouse tyrosinase-related protein 1 (*TRP-1*, also known as *gp75*) is a genetically engineered viral vector vaccine designed to provoke immune responses against melanoma. The drug consists of a vaccinia virus engineered to express the mouse TRP-1 antigen, a melanocyte differentiation protein found in both normal and malignant melanocytes. Immunization with this vector induces immune self-reactivity, leading to depigmentation (vitiligo) and effective immune protection against melanoma in preclinical mouse models. The antitumor effect is mediated primarily via CD4+ T lymphocyte-dependent immune activation and robust anti-TRP-1 antibody production, suggesting a mechanism of action involving both cellular and humoral arms of the immune system. The vaccine was shown in murine models to protect against lethal challenge with B16 melanoma and induce rejection of tumors by targeting TRP-1-positive cells[3].
