Overview

This drug is an ultraviolet (UV)-inactivated, non-replicating recombinant vaccinia virus vaccine developed by the University Hospital of Basel for the treatment of metastatic melanoma. The viral vector is engineered to co-express five specific transgenes from a single construct: three melanoma-associated tumor antigens (endoplasmic reticulum-targeted HLA-A0201-restricted Melan-A/MART-1, gp100, and tyrosinase epitopes) and two costimulatory molecules (CD80 and CD86). By delivering these components directly into lymph nodes via intranodal administration, the vaccine aims to enhance the presentation of tumor antigens to the immune system and provide necessary costimulatory signals to induce a potent cytotoxic T lymphocyte (CTL) response against melanoma cells. In clinical trials, it has been evaluated in a prime-boost strategy alongside synthetic melanoma-associated epitopes and granulocyte-macrophage colony-stimulating factor (GM-CSF) as a systemic adjuvant.
