Overview

The recombinant vesicular stomatitis virus human immunodeficiency virus vaccine (rVSV-HIV) is a live-attenuated, replication-competent viral vector vaccine candidate designed to prevent HIV-1 infection. Developed initially by Wyeth Research (now part of Pfizer) in collaboration with the HIV Vaccine Trials Network (HVTN), the vaccine utilizes a modified vesicular stomatitis virus (VSV) platform. To ensure safety and reduce neurovirulence, the VSV vector is attenuated through genetic modifications such as translocation of the N gene and truncation of the G protein. The vector is engineered to express HIV-1 specific antigens, primarily the Gag and Env proteins, to stimulate both robust T-cell mediated immunity and neutralizing antibody responses. Clinical evaluation, including the HVTN 090 study, focused on the safety and immunogenicity of various attenuated constructs in human subjects.
