Shingrix is a non-live, recombinant subunit vaccine developed by GSK for the prevention of herpes zoster (shingles) and postherpetic neuralgia in adults. It consists of the Varicella Zoster Virus (VZV) glycoprotein E (gE) antigen produced in Chinese Hamster Ovary cells, combined with the AS01B adjuvant system. The AS01B adjuvant is a liposome-based system containing two immunostimulants: 3-O-desacyl-4'-monophosphoryl lipid A (MPL) and QS-21. This combination is designed to enhance the antigen-specific cell-mediated immune response, which is critical for controlling the reactivation of VZV. The vaccine is typically administered as a two-dose series. The University of Wisconsin, Madison has been involved in clinical research evaluating the vaccine's safety and immunogenicity in specific populations, such as patients with inflammatory bowel disease (IBD) receiving biologic or JAK inhibitor therapies.
