Reconval K1 is a topical supportive care formulation developed by Pharmadab to manage skin toxicities, specifically acneiform rashes and lesions, induced by chemotherapy or targeted cancer therapies such as Epidermal Growth Factor Receptor (EGFR) inhibitors (e.g., cetuximab, panitumumab). The formulation contains 0.1% vitamin K1 (phylloquinone) in a moisturizing base that includes urea. Its mechanism of action involves the local activation of EGFR signaling in the skin, which helps counteract the inhibitory effects of systemic anti-EGFR treatments on dermal tissues without interfering with the systemic anti-tumor efficacy. Clinical data, including the randomized Phase II EVITA trial, have demonstrated that prophylactic use of Reconval K1 can reduce the incidence and severity of cetuximab-induced skin toxicity, particularly in women, thereby improving patient quality of life and treatment adherence. It is available as both a cream and a spray and is often marketed as a medical device for supportive care.
