Overview

recPRAME is a recombinant fusion protein comprising the full-length PRAME (Preferentially Expressed Antigen in Melanoma) tumor antigen. It is produced in *Escherichia coli* and includes an N-terminal tripeptide, a segment of *Haemophilus influenzae* protein D for enhanced immunogenicity, the 509-amino acid human PRAME sequence, and a C-terminal hexahistidine tag for purification[1]. recPRAME is developed as part of an antigen-specific cancer immunotherapeutic (ASCI), typically combined with the proprietary adjuvant AS15. The drug aims to stimulate the patient’s immune system to recognize and attack tumor cells expressing the PRAME antigen. Preclinical studies have shown that repeated injections induce robust antibody and T-cell responses against PRAME-expressing tumors[4]. Clinical development has focused on its use as adjuvant therapy in resected non-small cell lung cancer (NSCLC) and melanoma[1][2][7]. The developer is GlaxoSmithKline.
