recPRAME + AS15 is a **therapeutic cancer vaccine** composed of recombinant **PRAME antigen** (Preferentially Expressed Antigen in Melanoma) combined with the **AS15 immunostimulant adjuvant system**. PRAME is a tumor-associated antigen that is overexpressed in a variety of cancers including melanoma and non-small cell lung cancer (NSCLC); it is minimally expressed in most normal adult tissues. AS15 is a proprietary adjuvant from GSK that includes 3-O-desacyl-4′-monophosphoryl lipid A (MPL), QS-21 (a saponin licensed from Agenus), and CpG 7909 oligodeoxynucleotide, delivered in a liposomal formulation. The combination is designed to trigger both humoral and cellular immune responses against PRAME-expressing tumor cells, primarily by inducing PRAME-specific CD4+ and CD8+ T cell responses and antibody production. Preclinical studies and early clinical trials have shown immunogenicity, antitumor activity, and a favorable safety profile. The drug has been primarily developed for adjuvant treatment of PRAME-positive cancers, particularly melanoma and NSCLC, in post-surgical and minimal residual disease settings[1][2][3][4][5].
