Refanezumab (GSK249320) is a humanized IgG1 monoclonal antibody that targets myelin-associated glycoprotein (MAG). It was developed by GlaxoSmithKline for the recovery of motor function after stroke and has been investigated in clinical trials for cerebrovascular accident and ischemic attack. Refanezumab binds to MAG, blocking its inhibitory effect on axonal regeneration, thereby promoting axonal growth and potentially aiding neurological recovery post-stroke. Preclinical studies demonstrated its ability to cross the blood-brain barrier and neutralize MAG-mediated inhibition of neurite outgrowth. Clinical studies showed it was generally well-tolerated with no major safety issues; common adverse events included headache and upper respiratory tract infection[1][3][5].
