Refoxy Pharma is developing a lead series of small molecule activators targeting the Forkhead box O3 (FOXO3) transcription factor for the treatment of Idiopathic Pulmonary Fibrosis (IPF). FOXO3 is a key regulator of cellular stress resistance, metabolism, and autophagy, and its activity is known to decline with age and in chronic diseases like IPF. In fibrotic lung tissue, reduced FOXO3 expression in fibroblasts is associated with increased myofibroblast activation and resistance to apoptosis. By pharmacologically enhancing FOXO3 activity, Refoxy's small molecules aim to restore homeostatic signaling, promote fibroblast apoptosis, and reduce the production of extracellular matrix components, thereby slowing or reversing the progression of lung fibrosis. The program is currently in the preclinical stage of development.
