RegaVax is a live-attenuated, recombinant vector vaccine candidate that uses the well-established yellow fever 17D vaccine virus backbone to express the SARS-CoV-2 spike protein, thereby inducing immunity against both yellow fever and COVID-19.[1][7][17][19][20] Developed at the Rega Institute for Medical Research (KU Leuven), the vaccine is engineered by inserting the genetic code for the coronavirus spike into the yellow fever vaccine genome, resulting in a single-dose construct that in preclinical hamster and macaque models elicited rapid, high-titer neutralizing antibodies, protected against SARS-CoV-2 infection and pneumonia, and simultaneously functions as a yellow fever vaccine.[1][7][15][17][20] The candidate is positioned as a platform technology for live-attenuated recombinant vaccines based on the yellow fever vector and has progressed through preclinical development with preparations for clinical trials.[1][2][11][14][19]
