Overview

ReGel is a proprietary, thermosensitive, biodegradable hydrogel drug delivery platform originally developed by MacroMed. It consists of a triblock copolymer of poly(lactide-co-glycolide) (PLGA) and polyethylene glycol (PEG). The system is unique because it exists as a low-viscosity liquid at temperatures below room temperature (e.g., 2–15°C) but undergoes a spontaneous phase transition to a water-insoluble gel at body temperature (37°C). This allows for the easy injection of a drug-polymer mixture, which then forms a localized depot for the sustained release of therapeutic agents, ranging from small molecules to proteins and peptides. The most prominent application was OncoGel, an injectable formulation of paclitaxel for the local treatment of solid tumors, which aimed to provide high local concentrations of the cytotoxic agent while minimizing systemic toxicity.
