Overview

REGN1400 is a fully human monoclonal antibody developed by Regeneron Pharmaceuticals that targets the Receptor tyrosine-protein kinase erbB-3 (ERBB3/HER3) receptor, a member of the epidermal growth factor receptor (EGFR) family. By binding to ErbB3, REGN1400 prevents neuregulin 1 ligand from activating the receptor, thereby inhibiting downstream phosphatidylinositol-3 kinase (PI3K)/Akt signaling. This mechanism leads to reduced cellular proliferation and differentiation in cancer cells. Preclinical studies demonstrated that dual inhibition of ErbB3 and EGFR enhances antitumor activity compared to targeting either alone. REGN1400 was investigated primarily for various types of cancer, including non-small cell lung cancer (NSCLC), colorectal cancer (CRC), and squamous cell carcinoma of the head and neck (SCCHN). Clinical development reached phase 1 trials but was subsequently discontinued for oncology indications[1][2][4][6].
