Overview

REGN1400 + cetuximab is an investigational combination therapy of two monoclonal antibodies: REGN1400, a fully human monoclonal antibody targeting receptor tyrosine-protein kinase erbB-3 (HER3/ERBB3), and cetuximab, a chimeric monoclonal antibody that blocks epidermal growth factor receptor (EGFR). The rationale for this combination stems from evidence that dual inhibition of HER3/ERBB3 and EGFR can provide enhanced antitumor activity, as HER3/ERBB3 may mediate resistance to EGFR blockade. REGN1400 inhibits ERBB3-mediated activation of the Akt pathway, while cetuximab blocks EGFR-mediated signaling, together blocking critical cell proliferation and survival signals in certain solid tumors, such as non-small cell lung cancer, colorectal cancer, and squamous cell carcinoma of the head and neck. This combination has been evaluated in early-phase clinical trials, primarily for patients with advanced cancers who progressed following prior EGFR inhibitor therapy[1][2][3][7].
