REGN17372 is an investigational bispecific monoclonal antibody developed by Regeneron Pharmaceuticals for the treatment of multiple myeloma. It is a costimulatory T-cell engager designed to simultaneously bind to G protein-coupled receptor class C group 5 member D (GPRC5D) on multiple myeloma cells and the CD28 receptor on T cells. By targeting CD28, REGN17372 provides a "Signal 2" costimulatory stimulus that is intended to enhance the activation, proliferation, and cytotoxic activity of T cells, particularly when used in conjunction with "Signal 1" agents such as CD3-bispecific antibodies (e.g., linvoseltamab). This combination approach aims to overcome T-cell exhaustion and improve the durability of responses in patients with relapsed or refractory multiple myeloma. It is currently being evaluated in Phase 1/2 clinical trials.
