Overview

REGN20423 is an investigational monoclonal antibody developed by Regeneron Pharmaceuticals that targets the interleukin-13 receptor subunit alpha 1 (IL-13Rα1). It is currently in Phase 1 clinical development for the treatment of atopic dermatitis. By binding to IL-13Rα1, REGN20423 inhibits the formation of the Type II IL-4/IL-13 receptor complex, which is a critical mediator of Type 2 inflammation. Unlike IL-4Rα antagonists (such as dupilumab) which block both Type I and Type II receptors, an IL-13Rα1 antagonist specifically targets the Type II complex, potentially offering a differentiated profile by sparing IL-4 signaling through the Type I receptor (IL-4Rα/γc) found on hematopoietic cells.
