Overview

REGN5054 is a bispecific monoclonal antibody developed by Regeneron Pharmaceuticals that targets Mucin 16 (MUC16) and the T-cell costimulatory receptor CD28. It is part of Regeneron's costimulatory bispecific platform, designed to provide a localized Signal 2 to T-cells within the tumor microenvironment. MUC16 is a high-molecular-weight glycoprotein frequently overexpressed in ovarian, endometrial, and fallopian tube cancers. By bridging MUC16-expressing tumor cells with CD28 on T-cells, REGN5054 enhances T-cell activation and proliferation when combined with a primary Signal 1 (such as that provided by a PD-1 inhibitor like cemiplimab or a CD3-targeting bispecific). This approach aims to maximize anti-tumor efficacy while minimizing the systemic inflammatory side effects often associated with systemic CD28 agonism.
