Overview

REGN5668 + cemiplimab is an investigational combination therapy currently in clinical trials for advanced epithelial ovarian cancer, fallopian tube cancer, and primary peritoneal cancer. REGN5668 is a human IgG4-based bispecific monoclonal antibody that binds both Mucin-16 (MUC16), a cell surface glycoprotein overexpressed in ovarian cancer, and CD28 on T cells, providing T cell costimulation to promote tumor-directed cytotoxicity. Cemiplimab is a human monoclonal antibody targeting programmed death-1 (PD-1), blocking PD-1 interaction with its ligands PD-L1 and PD-L2, thereby enhancing T cell immune responses against tumors. REGN5668 demonstrated increased preclinical antitumor activity when combined with PD-1 inhibition (cemiplimab) relative to monotherapy. The combination aims to increase T cell-mediated killing of MUC16+ tumor cells by simultaneously providing costimulatory and checkpoint blockade signals[3][1][7][4][5].
