Overview

REGN5668 + ubamatamab is an investigational combination of two bispecific monoclonal antibodies developed for the treatment of ovarian cancer and other MUC16-expressing gynecologic malignancies. Both agents are designed to engage T cells for enhanced anti-tumor activity: - **REGN5668** is a human IgG4-based bispecific antibody that binds to MUC16 (a cell surface glycoprotein overexpressed in ovarian cancer) and CD28 (a T-cell costimulatory receptor), providing a costimulatory signal to T-cells in the tumor microenvironment[4]. - **Ubamatamab (REGN4018)** is a bispecific antibody targeting MUC16 and CD3, promoting direct T cell-mediated cytotoxicity against cancer cells expressing MUC16[2][4]. Their combination is hypothesized to synergistically stimulate T-cell activation and cytotoxicity, leading to improved anti-tumor immune responses compared to single-agent therapy. The combination is currently being evaluated in phase 1/2 trials for safety, tolerability, pharmacokinetics, and preliminary efficacy in patients with recurrent ovarian cancer and related malignancies[1][4][5][8][9].
