Overview

REGN5668 + ubamatamab + sarilumab is an experimental combination of three monoclonal antibodies with immunotherapeutic mechanisms, investigated primarily for ovarian cancer and related gynecologic malignancies. REGN5668 is a bispecific antibody that targets MUC16 (CA125) on tumor cells and CD28 on T cells to co-stimulate T-cell cytotoxicity. Ubamatamab (also known as REGN4018) is a bispecific antibody targeting MUC16 and CD3, facilitating T-cell mediated killing of MUC16-positive cancer cells. Sarilumab is a monoclonal antibody targeting the interleukin-6 receptor (IL-6R) and is approved for rheumatoid arthritis. While clinical trials have evaluated REGN5668 with other agents (cemiplimab or REGN4018), there are currently no major studies combining all three agents (REGN5668, ubamatamab, and sarilumab) together; this combination may be hypothetical or under preclinical consideration. REGN5668 and ubamatamab are both developed by Regeneron, as is sarilumab, and all are designed to stimulate and modulate the immune response against cancer, especially in MUC16-positive recalcitrant ovarian cancers[1][2][3][6].
