REGN5837 is a hinge-stabilized, human immunoglobulin G4 (IgG4) bispecific monoclonal antibody developed by Regeneron Pharmaceuticals. It targets both CD22, a tumor-associated antigen expressed on B cells, and CD28, a co-stimulatory molecule on T cells. By binding to these targets, REGN5837 provides a co-stimulatory signal (signal 2) to T cells in the presence of CD22-expressing tumor cells. This mechanism is designed to enhance T cell activation and cytotoxicity against malignant B cells when used in combination with odronextamab (a CD20xCD3 bispecific antibody that provides signal 1). The drug is being investigated primarily for relapsed or refractory aggressive B-cell non-Hodgkin lymphomas and has shown potential immunostimulating and antineoplastic activities in preclinical models[1][3][5][8].
