REGN6641 (also known as SAR444419) is an investigational bispecific monoclonal antibody co-developed by Regeneron Pharmaceuticals and Sanofi for the treatment of Type 2 inflammatory diseases. It is designed to simultaneously target and inhibit two key receptors: the interleukin-4 receptor alpha (IL-4Rα) and the interleukin-33 receptor (ST2, also known as IL-1RL1). By blocking IL-4Rα, REGN6641 inhibits the signaling of both IL-4 and IL-13, which are central drivers of allergic and inflammatory conditions. Simultaneously, by antagonizing ST2, it prevents the action of IL-33, an upstream alarmin cytokine that triggers and amplifies potent inflammatory responses. This dual-targeting approach is intended to provide more comprehensive suppression of the multiple pathways contributing to chronic Type 2 inflammation compared to monotherapies. REGN6641 is currently in early-stage clinical development, primarily being evaluated for atopic dermatitis.
