Overview

REGN7945 is an investigational bispecific monoclonal antibody developed by Regeneron Pharmaceuticals. It targets both CD38 and CD28, functioning as a costimulatory bispecific antibody designed to enhance T cell activation in the context of cancer immunotherapy. By binding to CD38 (highly expressed on multiple myeloma cells) and providing targeted costimulation through CD28 on T cells, REGN7945 aims to augment the anti-tumor activity of other T cell-engaging therapies such as linvoseltamab (a BCMAxCD3 bispecific antibody). Preclinical studies have shown that combining REGN7945 with linvoseltamab enhances T cell activation, cytokine production, cytotoxicity, and anti-tumor efficacy in models of relapsed/refractory multiple myeloma. The drug is currently being evaluated in a first-in-human phase 1/2 clinical trial for adults with relapsed or refractory multiple myeloma[2][7][8].
