REGN7999 is a monoclonal antibody developed by Regeneron that targets and inhibits transmembrane protease serine 6 (TMPRSS6). By blocking TMPRSS6, REGN7999 prevents the cleavage of hemojuvelin (HJV), which enhances BMP6-HJV signaling and increases serum hepcidin levels. Elevated hepcidin reduces iron absorption and mobilization, leading to decreased serum iron and liver iron stores. This mechanism is being investigated as a treatment for iron overload conditions, particularly in patients with non-transfusion dependent beta-thalassemia. Preclinical studies have shown that REGN7999 can significantly reduce liver iron content and improve red blood cell health in animal models. Clinical trials are ongoing to assess its safety, tolerability, and efficacy in humans[1][3][5][6][8].
