Regulatory Donor-Derived Dendritic Cells (DCreg) are a type of cellular therapy derived from donor bone marrow cells that are processed to exhibit immunoregulatory properties. These cells are designed to promote transplant tolerance by down-regulating alloimmune responses. DCreg express a tolerogenic gene transcriptional profile, high cell surface programed death ligand-1 (PD-L1):CD86 ratios, high IL-10 production with no IL-12 production, and demonstrate poor ability to stimulate allogeneic T cell proliferation. They are resistant to maturation and to becoming immunogenic after exposure to pro-inflammatory stimuli. In transplantation, DCreg are typically infused into recipients before organ transplantation to help reduce dependency on immunosuppressive drugs and potentially promote operational tolerance.
