Relÿva is a topical small molecule formulation of rofecoxib, a selective cyclooxygenase-2 (COX-2) inhibitor, being developed by BriOri BioTech for the treatment of osteoarthritis-related pain in the knee and joints. Rofecoxib was previously marketed as the oral drug Vioxx but was withdrawn from the market due to cardiovascular risks associated with systemic exposure. Relÿva is designed to provide potent, 24-hour, non-addictive pain relief by delivering the medication directly to the site of inflammation, thereby achieving lower systemic exposure and an improved safety profile compared to oral nonsteroidal anti-inflammatory drugs (NSAIDs). As of 2022, the drug has demonstrated safety and efficacy in preclinical models and is being positioned for clinical advancement.
