Relatlimab is a human IgG4 monoclonal antibody and immune checkpoint inhibitor that targets lymphocyte activation gene-3 (LAG-3), an inhibitory receptor on T cells. By blocking LAG-3, relatlimab restores T-cell function and enhances anti-tumor immune responses. It is primarily used in combination with nivolumab, a PD-1 inhibitor, as the fixed-dose product Opdualag for the treatment of unresectable or metastatic melanoma in adults and pediatric patients aged 12 years and older. Developed by Bristol Myers Squibb, it was approved by the FDA in March 2022 as the first LAG-3-blocking combination therapy for melanoma[1][4][5][8].
