This drug is a combination of three agents: **relatlimab**, a monoclonal antibody targeting lymphocyte activation gene-3 (LAG-3); **nivolumab**, a monoclonal antibody targeting programmed death-1 (PD-1); and **linrodostat**, an investigational small molecule inhibitor of indoleamine 2,3-dioxygenase 1 (IDO1). Relatlimab and nivolumab potentiate immune responses by inhibiting checkpoint mechanisms that suppress T-cell activity, enhancing the immune system’s ability to attack tumor cells. Linrodostat is designed to suppress tumor-mediated immunosuppression by blocking IDO1, an enzyme involved in tryptophan catabolism and immune tolerance within the tumor microenvironment. This multi-agent combination is under investigation as an immunotherapeutic approach to treat solid tumors, with potential primary indications in cancer, particularly melanoma and other checkpoint blockade-responsive malignancies.
