Reliaject

Reliaject is a branded formulation of methotrexate, a folate analogue antimetabolite, provided as a solution for subcutaneous injection in a pre-filled autoinjector. Developed and marketed by Accord Healthcare, it is primarily used as a disease-modifying antirheumatic drug (DMARD) for the treatment of severe active rheumatoid arthritis, polyarthritic forms of severe juvenile idiopathic arthritis, and severe recalcitrant psoriasis. Its mechanism of action involves the competitive inhibition of dihydrofolate reductase (DHFR), which prevents the formation of tetrahydrofolate required for purine and pyrimidine synthesis, thereby inhibiting DNA replication in rapidly dividing cells. In inflammatory conditions, methotrexate also inhibits aminoimidazole carboxamide ribonucleotide transformylase (ATIC), leading to increased extracellular adenosine levels which mediate potent anti-inflammatory effects.
