Overview

Rember is a small molecule drug developed as a tau aggregation inhibitor for the treatment of Alzheimer's disease. It is based on the chemical structure of methylene blue and specifically targets the aggregation of tau protein in neurofibrillary tangles—a hallmark pathology in Alzheimer’s disease. Unlike traditional Alzheimer’s therapies that target amyloid plaques or provide symptomatic relief via cholinesterase inhibition, rember aims to slow disease progression by inhibiting tau protein aggregation. The original compound was later modified and improved into leuco-methylthioninium-bis(hydromethanesulfonate) (LMTM), also known as TRx-0237[4][2][5]. Clinical trials have shown mixed results; while early phase II data suggested significant slowing of cognitive decline compared to standard treatments like donepezil (Aricept), subsequent phase III trials did not meet their primary endpoints overall but showed some benefit in patients not taking other Alzheimer's medications[4][2]. Side effects include gastrointestinal symptoms and discoloration of urine due to its dye properties[4].
