Overview

REN-1654 is a small molecule drug that was developed as an inhibitor of tumor necrosis factor alpha (TNF-alpha) and p38 mitogen-activated protein kinase (p38 MAPK). It was investigated for its potential neuroprotective and anti-inflammatory effects in several indications, including neuropathic pain conditions such as post-herpetic neuralgia, sciatica, lumbosacral radiculopathy, cognitive impairment associated with AIDS dementia complex and other cognitive dysfunctions. The drug modulates the JNK signaling pathway and the MAPK/ERK signaling pathway. Despite reaching Phase 2 clinical trials for multiple indications, development of REN-1654 was discontinued after it failed to demonstrate significant efficacy in mid-stage studies[2][5][7][8].
