RENB-DC-11 is a next-generation, genetically engineered allogeneic dendritic cell therapy developed by Renovaro Biosciences for the treatment of solid tumors, with an initial focus on pancreatic cancer. This therapy uses CD34+ hematopoietic stem cells from healthy donors, which are engineered to express the immune-modulating molecules CD93, CD40L, and CXCL13. The modifications aim to enhance the dendritic cells’ ability to activate both CD4+ helper T cells and CD8+ cytotoxic T cells, as well as NK cells, resulting in potent anti-tumor immune responses. Preclinical studies in humanized mouse models have demonstrated up to 81.7% reduction in pancreatic tumor size, significant immune activation, antigen-specific tumor cell killing, and a lack of metastases. The approach is designed as an “off-the-shelf” therapy utilizing scalable and cryopreservable manufacturing. Development plans include expansion into additional solid tumor types and clinical trials starting with pancreatic cancer.
