RENCA mitochondrial lysate dendritic cell vaccine is an experimental cancer immunotherapy investigated in preclinical research for the treatment of renal cell carcinoma. The vaccine is composed of autologous or syngeneic dendritic cells pulsed with an enriched mitochondrial protein lysate derived from RENCA (murine renal adenocarcinoma) tumor cells. This approach targets tumor-associated mitochondrial antigens (TAMAs), which are potentially immunogenic due to somatic mitochondrial DNA (mtDNA) mutations frequently found in various solid tumors. In murine models, the vaccine has demonstrated the ability to elicit a robust CD8+ cytotoxic T cell response, providing durable protection against tumor challenge in both prophylactic and therapeutic settings. Research has identified specific mutated mitochondrial proteins, such as Cytochrome c oxidase subunit 1 (COX1), as key immunogenic targets within the lysate that drive the antitumor immune response.
