The RENCA tumor cell vaccine is an experimental whole-cell cancer immunotherapy primarily utilized in preclinical research for renal cell carcinoma (RCC). It is based on the murine RENCA (renal adenocarcinoma) cell line and typically consists of irradiated tumor cells. These vaccines are often genetically modified to secrete immunostimulatory cytokines, most notably granulocyte-macrophage colony-stimulating factor (GM-CSF), or other factors like IL-6 or Hyper-IL-6, to enhance their immunogenicity. The vaccine functions by presenting a broad spectrum of tumor-associated antigens to the host immune system, while the secreted cytokines (like GM-CSF) recruit and mature dendritic cells at the vaccination site. This process primes tumor-specific CD4+ and CD8+ T-lymphocytes, leading to systemic antitumor immunity. It is a standard model for studying cell-based vaccination strategies in syngeneic BALB/c mice.
