REP 2055 is a nucleic acid polymer (NAP), specifically a synthetic, amphipathic, oligonucleotide composed of polycytidine and approximately 40 nucleotides in length. It was developed as an antiviral agent targeting hepatitis B virus (HBV) and has also shown activity against hepatitis C virus (HCV) and duck hepatitis B virus (DHBV). Its primary mechanism is to inhibit the release of hepatitis B surface antigen (HBsAg) from infected hepatocytes, which is believed to help restore immune control over chronic HBV infection. Unlike some other NAPs, it does not contain CpG motifs and does not directly stimulate the immune system. In preclinical models and early clinical studies, it demonstrated both prophylactic and therapeutic effects by reducing viral markers in serum and promoting sustained virologic responses after treatment cessation[1][2][3][5][6].
