The replicon RNA COVID-19 vaccine is a self-amplifying RNA (saRNA) vaccine candidate being developed for the prevention of COVID-19. Unlike conventional mRNA vaccines, saRNA utilizes a replicase complex (typically derived from alphaviruses) that allows the RNA to self-amplify within the host cell after administration. This mechanism leads to sustained and high-level expression of the encoded antigen—in this case, the SARS-CoV-2 spike protein—which can potentially achieve robust immune responses at significantly lower doses than standard mRNA vaccines. The Research Institute for Microbial Diseases (BIKEN Foundation) and VLP Therapeutics Japan entered into a Memorandum of Understanding in September 2023 to collaborate on the development and manufacturing of this vaccine, which is currently in the preclinical stage.
