Overview

reRNA-Melanoma is an investigational self-amplifying RNA (saRNA) cancer vaccine designed for the treatment of patients with unresectable stage III or IV melanoma. Developed by The First Affiliated Hospital of Nanyang Medical College, the vaccine utilizes a replicating RNA (reRNA) platform, which typically incorporates viral replicon components (such as those from alphaviruses) to enable the RNA to self-duplicate within host cells. This mechanism is intended to provide higher and more sustained levels of tumor-associated antigen expression compared to conventional non-replicating mRNA vaccines, potentially eliciting a more robust and durable T-cell-mediated anti-tumor immune response. The vaccine is currently being evaluated in a Phase 1 clinical trial in China, both as a monotherapy and in combination with the PD-1 inhibitor toripalimab, to assess safety, tolerability, immunogenicity, and preliminary efficacy in patients who have failed at least one prior systemic therapy.
