RESET33 is a potent, synthetic small molecule agonist of the nuclear receptors REV-ERBα (NR1D1) and REV-ERBβ (NR1D2). These receptors are integral components of the mammalian circadian clock, functioning as transcriptional repressors that regulate various physiological processes, including metabolism, inflammation, and sleep-wake cycles. Developed by Reset Therapeutics, RESET33 was designed to treat metabolic disorders such as type 2 diabetes and obesity by synchronizing or modulating the molecular clock's influence on glucose and lipid homeostasis. In preclinical models, activation of REV-ERB by RESET33 has been shown to reduce fat mass, improve plasma lipid profiles, and enhance insulin sensitivity.
