Overview

Resolvin D2 is a specialized pro-resolving lipid mediator of the resolvin family, biosynthesized from the omega-3 fatty acid docosahexaenoic acid (DHA) via sequential oxygenation by 15- and 5-lipoxygenase. It exhibits potent anti-inflammatory and pro-resolving activity, promoting the resolution of inflammation, modulating immune cell function, and facilitating tissue regeneration. Mechanistically, resolvin D2 activates the G protein-coupled receptor GPR18 (also known as DRV2), leading to a switch of macrophages toward an anti-inflammatory phenotype, increased secretion of pro-myogenic factors, and direct enhancement of myogenic cell differentiation. Preclinical models demonstrate activity in reducing inflammation, promoting muscle regeneration, and alleviating inflammatory pain. Resolvin D2 is under investigation as a therapeutic candidate for conditions driven by chronic inflammation, including Duchenne muscular dystrophy and pain syndromes[1][2][3][9].
