Restoret is an investigational, potentially first-in-class tetravalent, tri-specific Wnt pathway agonist antibody developed for the treatment of retinal diseases, primarily diabetic macular edema (DME) and neovascular age-related macular degeneration (NVAMD). It is designed to restore and maintain the blood-retinal barrier by activating the Wnt signaling pathway in the retina, thereby reducing vascular leakage. Unlike anti-VEGF therapies that target vascular endothelial growth factor to reduce abnormal blood vessel growth and leakage, Restoret works through a novel mechanism as a mimetic of norrin—the natural Wnt ligand in the eye—offering an alternative approach for patients with suboptimal response to current standard-of-care treatments. The drug is administered via intravitreal injection. Clinical trials have shown promising safety and efficacy results, with improvements in visual acuity and reduction in retinal thickness observed in early-phase studies[1][2][3][4][5][6][7].
