Overview

Rethymic (allogeneic cultured thymus tissue-agdc) is a regenerative medicine therapy consisting of processed thymus tissue slices, developed by Enzyvant (a subsidiary of Sumitomo Pharma). It is the first FDA-approved treatment for immune reconstitution in pediatric patients with congenital athymia, a condition most commonly associated with Complete DiGeorge Syndrome (cDGS). The therapy works by providing a functional thymic microenvironment that supports the maturation of host-derived T-cell progenitors, which are otherwise unable to develop due to the absence of a thymus. In patients with cDGS who also present with hypoparathyroidism, Sumitomo is evaluating a combination approach (NCT04293159) involving the co-transplantation of cultured parathyroid tissue alongside the thymus tissue. This investigational procedure aims to simultaneously address the life-threatening immune deficiency and the chronic calcium regulation issues by providing a source of parathyroid hormone (PTH).
