Overview

Retifanlimab + INCB001158 is a combination therapy under investigation for the treatment of advanced solid tumors. Retifanlimab is a humanized IgG4 kappa monoclonal antibody that targets programmed cell death protein-1 (PD-1), blocking its interaction with PD-L1 and PD-L2, thereby enhancing T-cell–mediated immune responses against cancer cells. It is approved as monotherapy for metastatic or recurrent locally advanced Merkel cell carcinoma under the brand name Zynyz[3][5][6]. INCB001158 is an oral small molecule inhibitor of arginase, an enzyme involved in immunosuppression within the tumor microenvironment. By inhibiting arginase, INCB001158 aims to reduce immunosuppressive metabolites and further promote anti-tumor immunity[1][2]. The combination has been studied in phase Ib/II trials to assess safety, tolerability, and preliminary efficacy in patients with advanced solid tumors[1][2].
