Overview

Retlirafusp alfa (SHR-1701) is a bifunctional fusion protein biologic that simultaneously targets programmed death-ligand 1 (PD-L1) and transforming growth factor-beta (TGF-β). Developed by Suzhou Suncadia Biopharmaceuticals in collaboration with Jiangsu Hengrui Medicine, it is designed to overcome immune evasion in the tumor microenvironment by blocking both the PD-1/PD-L1 checkpoint and the immunosuppressive TGF-β signaling pathway. In the context of rectal cancer, it is being investigated as a perioperative treatment for resectable locally advanced rectal cancer (LARC) in combination with chemoradiotherapy and XELOX chemotherapy. This dual-targeting strategy aims to enhance anti-tumor immune responses more effectively than single-agent checkpoint inhibition.
