Retlirafusp alpha (SHR-1701) is a bifunctional fusion protein developed by Jiangsu Hengrui Pharmaceuticals. It consists of a humanized monoclonal antibody targeting programmed death-ligand 1 (PD-L1) fused to the extracellular domain of the human transforming growth factor-beta receptor type II (TGFBR2). This dual-action molecule is designed to simultaneously inhibit the PD-1/PD-L1 checkpoint pathway and sequester TGF-beta within the tumor microenvironment. By blocking these two major immunosuppressive pathways, retlirafusp alpha aims to restore anti-tumor immune responses and overcome resistance to standard checkpoint inhibitors. It is currently being evaluated in various clinical trials, particularly for gastrointestinal cancers and non-small cell lung cancer.
